Characterization of abalone Haliotis tuberculata-Vibrio harveyi interactions in gill primary cultures ArchiMer

The decline of European abalone Haliotis tuberculata populations has been associated with various pathogens including bacteria of the genus Vibrio. Following the summer mortality outbreaks reported in France between 1998 and 2000, Vibrio harveyi strains were isolated from moribund abalones, allowing in vivo and in vitro studies on the interactions between abalone H. tuberculata and V. harveyi. This work reports the development of primary cell cultures from abalone gill tissue, a target tissue for bacterial colonisation, and their use for in vitro study of host cell—V. harveyi interactions. Development of a mreB-targeted real-time PCR method for the quantitative detection of Vibrio harveyi in seawater and biofilm from aquaculture systems ArchiMer

Vibrio harveyi is a particularly problematic Gram-negative bacterium because it can form biofilms on aquaculture facility surfaces, leading to resistance of bacteria against antibiotics and water sanitizers. A SYBR Green I quantitative real-time PCR method was developed to detect V. harveyi directly from environmental samples, including seawater and biofilm. Ecophysiological studies on luminous bacteria associated with marine gastropods. ArchiMer

Information available on the association of luminous bacteria with marine invertebrates is limited. Hence qualitative and quantitative investigations were undertaken with a view to understand the distribution of luminous microflora in relation to the environmental factors and also in the gut of two marine gastropods, Aplysia benedicti Eliot and Bullia tranquebarica (Roding) from Porto Novo (11 degree 29'N, 79 degree 46'E) waters of the East coast of India. El empleo de probióticos para el control de enfermedades en la acuicultura intensiva es una alternativa al uso de antibióticos. En este trabajo se aisló la cepa bacteriana IC1 a partir de una fermentación acido láctica de col y se identificó por técnicas moleculares y convencionales como Lactobacillus sp. Se determinó la actividad antagónica in vitro de IC1 contra cepas patrones. Effect of combined function of temperature and water activity on the growth of Vibrio harveyi BJM

Vibrio harveyi is considered as a causative agent of the systemic disease, vibriosis, which occurs in many biological fields. The effects of temperatures (12.9-27.1 ºC) and water activity (NaCl% 0.6%-3.4%) on V. harveyi were investigated. The behavior and growth characteristics of V. harveyi was studied and modeled. Growth curves were fitted by using Gompertz and Baranyi models, and the Baranyi model showed a better fittness. Then, the maximum growth rates (µmax) and lag phase durations (LPD, λ) obtained from both Gompertz and Baranyi model were modeled as a combination function of temperature and water activity using the response surface and Arrhenius-Davey models for secondary model. Since 1997, populations of the European abalone Haliotis tuberculata suffer mass mortalities attributed to the bacterium Vibrio harveyi. These mortalities occur at the spawning season, when the abalone immune system is depressed, and when temperatures exceed 17 °C, leading to favorable conditions for V. harveyi proliferation. In order to identify mechanisms of disease resistance, experimental successive infections were carried out on two geographically distinct populations: one that has suffered recurrent mortalities (Saint-Malo) and one that has not been impacted by the disease (Molène). Abalone aquaculture only recently developed in western Europe, but already it has to face numerous pathologies. Since 1998, the natural and farm stocks of Haliotis tuberculata suffer regularly high mortalities, mainly due to the pathogenic bacterium, Vibrio harveyi. The vibriosis of the European abalone, in particular its macroscopic signs, and the conditions necessary for its development have been studied in detail by field and laboratory experiments. Molecular variations in Vibrio alginolyticus and V. harveyi in shrimp-farming systems upon stress BJM

A study was performed to investigate the genomic variations in the shrimp farm isolates of Vibrio alginolyticus and V. harveyi when the isolates were subjected to environmental stress. Samples of shrimps, water and sediment were collected from Southern Indian coastal shrimp farms. Vibrio isolates were biochemically identified and confirmed using 16S rDNA and gyrB gene specific PCR. The bacterial strains were genotyped by PCR fingerprinting using GTG(5) and IS (Insertion Sequence) primers. The autoinducer AI-2 has been proposed to serve as a ‘universal signal’ for interspecies communication. This study aimed to evaluate the capability of Enterococcus faecium, Enterococcus faecalis, and Bacillus cereus strains isolated from ricotta processing to produce quorum sensing signalling molecules (AI-2). Sponges are a rich source for investigation of bioactive small molecules. They have been mostly investigated for the search of new pharmacological models or therapeutic agents for the treatment of human diseases. Micro-organisms can also represent a virulent pathogen for marine invertebrates such as sponges, which need to protect themselves against these microbes. Seasonal variation in the antivibrio activity of two organic extracts from two red seaweed: Palmaria palmata and the introduced Grateloupia turuturu against the abalone pathogen Vibrio harveyi ArchiMer

The wide polarity range and highly polar compounds of two selected red seaweed, Grateloupia turuturu and Palmaria palmata were extracted using two different types of solvent, dichloromethane/methanol and methanol/water. Monthly in vitro antibacterial activities were studied using the microplate method against the marine bacteria Vibrio harveyi strain ORM4, known to infect abalone. Inhibition, slowdown and delay of Vibrio harveyi growth were investigated. Polar compounds of seaweed showed an activity against the abalone pathogen. The best activity was recorded from P. palmata collected in spring, with an inhibition of 7.9% of the bacterial growth. Preliminary 1H NMR profiles identified the differences between the extracts.

The objective of this work was to assess the time-related action of probiotic Lactobacillus plantarum in the bacterial microbiota of the digestive tract of Litopenaeus vannamei, and the relation of total haemocyte count and serum phenol oxidase activity of shrimp challenged with Vibrio harveyi. Shrimps were fed with a probiotic-supplemented diet, for eight days, then shifted to a commercial diet. Shrimps fed only with the commercial diet served as control.
